East Sussex County Council will close part of the A259 Wallsend Road, including the level crossing, on Sunday, May 25.
The stretch will be shut between 7am and 4pm to allow works on the railway line.
Councillor Terry Swales raised the matter at a Pevensey Parish Council meeting recently.
Cllr Swales said, "It is the Bank Holiday weekend and concerns have been raised by residents."
Cllr Maurice Gilbert said, "To cut a village off on a Bank Holiday weekend is a concern. It should be closed during the night and reopened in the morning if possible."
The road was also shut during the early hours of Sunday (April 6). ESCC say these scheduled closures will enable Network Rail contractors to carry out essential civil engineering works on railway installations.
Access for pedestrians should not be affected.
